Customer planned to use the Anti-SLC12A3 antibody (HPA028748) for IHC in human kidney samples and asked if it is the distal kidney tubules or the collecting ducts that are stained by the antibody?

Atlas Antibodies immunohistochemistry QC team performed IHC staining of kidney sections displaying both the kidney medulla and kidney cortex. The existing images on the Human Protein Atlas for the Anti-SLC12A3 antibody (HPA028748) displays tissue cores from the cortex and not the medulla part of the kidney. Distal kidney tubules are present in the cortex and not in the medulla whereas the collecting ducts could be present both in cortex and medulla. In order to make sure if collecting ducts or distal tubules were stained, a section containing both the medulla and cortex was stained with the Anti-SLC12A3 antibody (HPA028748).

Images show staining of tubules in the cortex part of the kidney and not in the medulla, which supports that the antibody targets distal tubules and not collecting ducts.
